Formal Definition

Physiologically based toxicokinetic, or alternatively referred to as physiologically based pharmacokinetic or biokinetic models, are quantitative descriptions of absorption, distribution, metabolism, and excretion (ADME, possibly including toxicity as ADMET) of synthetic or natural chemical substances in humans and other animal species. PBTK models are increasingly being used as an effective tool for designing toxicology experiments and for conducting extrapolations essential for risk assessments (e.g., in pharmaceutical research and medicine development, and in health risk assessment for cosmetics or general chemicals).
